چکیده

Many anticancer drugs exert their cytotoxicity by inhibiting DNA synthesis and cell replication. However, side effects such as bone marrow suppression, gastrointestinal toxicity, and renal damage remain to be addressed (Peterson et al., 1992). One avenue to developing clinically applicable chemotherapeutic agents with fewer complications is to screen traditional medicinal plants, which have been used for thousands of years with a very low number of side effects, for their anticancer activity (Lee et al., 2007; Park et al., 2008; Ricci et al., 2010). Chios mastic gum (CMG) is a white resin obtained from the trunk and leaves of Pistacia lentiscus var. Chia. Mastic gum and its essential oils are natural antimicrobial agents that have been used extensively in Mediterranean and Middle Eastern countries, both as a dietary supplement and herbal remedy, since antiquity (Loizou et al., 2009). Mastic gum is known to have antioxidant (Abdel-Rahman and Soad, 1975), anti-bacterial (Magiatis et al., 1999; Koutsoudaki et al., 2005), cosmetic, and teeth-cleaning properties (Wellmann, 1907), as well as anti-Helicobacter pylori properties (Huwez et al., 1998; Marone et al., 2001). Recent studies demonstrate that CMG induces apoptosis of human colon cancer cells through caspase-dependent pathways in vitro and cell growth inhibition of prostate cancer cells (Balan et al., 2005; Balan et al., 2007), as well as inhibiting the proliferation of androgen-independent prostate cancer, apparently through modulation of the

Induction of apoptosis in human colon cancer HCT116 cells treated with an extract of the plant product, Chios mastic gum.

A hexane extract of the plant product Chios mastic gum (He-CMG) is demonstrated to kill human colon cancer cells in vitro via the process of anoikis. Specifically, the sequence of events includes He-CMG-induced GI-arrest of the cells, detachment of the cells from the substrate and subsequent apoptosis. Anoikis is dependent on the concentration and duration of treatment with He-CMG. Presence of ...

Anti-inflammatory activity of Chios mastic gum is associated with inhibition of TNF-alpha induced oxidative stress

BACKGROUND Gum of Chios mastic (Pistacia lentiscus var. chia) is a natural antimicrobial agent that has found extensive use in pharmaceutical products and as a nutritional supplement. The molecular mechanisms of its anti-inflammatory activity, however, are not clear. In this work, the potential role of antioxidant activity of Chios mastic gum has been evaluated. METHODS Scavenging of superoxi...

Suppressive Effect of Constructed shRNAs against Apollon Induces Apoptosis and Growth Inhibition in the HeLa Cell Line

Background: Cervical cancer is the second most common female cancer worldwide. Inhibitors of apoptosis proteins (IAPs) block apoptosis; therefore, therapeutic strategies targeting IAPs have attracted the interest of researchers in recent years. Apollon, a member of IAPs, inhibits apoptosis and cell death. RNA interference is a pathway in which small interfering RNA (siRNA) or shRNA (short hairp...

A mastic gum extract induces suppression of growth of human colorectal tumor xenografts in immunodeficient mice.

BACKGROUND We recently reported that ethanol and hexane extracts of the plant product, mastic gum (MG), contain constituents which can induce p53- and p21-independent G1-phase arrest followed by apoptosis of human HCT116 colon cancer cells in vitro. Herein, we extended these studies to investigate the in vivo anticancer activity of the hexane extract of MG (He-MG) against human colon tumor. The...
